The big issue with cutting the City - Stafford section of the 375 is the fact that residents are being provided with an inferior service by a modified 369 (same frequency, forced transfer).

This, however, is debatable. I used a 375 from Herschel Street one time I was going out to Stafford City - god the trip through the City/Valley was slow as hell! It felt like it took us 20 minutes to get to RBWH, and this was at 10am on a Wednesday morning! (Looked at the timetable - and it is timetabled as 20 minutes). This slow trip through the City stuck in my mind so much that I got off and boarded a 333 to take me to the city! In fact, you could probably offset the transfer penalty just by co-ordinating a 369 with a 333 service. The 375 takes twice as long (additional 8 mins) to get to RBWH from the City (Adelaide St Stop 17/KGSBS)!

From my observations, a lot of passengers get off in the Gordon Park area (on my particular service, many seemed to be grannies). In this area, the bus also suffers from 'too many bus stops too close together'. Hence, 369 would absolutely need to go into that area. This is getting off topic, but the diversion of the 369 into Gordon Park could risk ruining the whole route as an effective cross-town bus, as would a Stafford City diversion.

Overall, I found the 375 to be painfully slow City - Stafford.
Is it really that hard to run frequent, reliable public transport?

Actually neither the 555 nor 250 existed pre-TransLink. As for BT, the wasteful duplication's been going on since the '80s/'90s or so, with more and more stops having been added to the Cityxpress routes.
